產(chǎn)品概述
IL-3是一種多效性的細(xì)胞因子,可刺激細(xì)胞的增殖分化并刺激多功能造血干細(xì)胞和譜系定向祖細(xì)胞的存活。表達(dá)IL-3的細(xì)胞有幾種造血和非造血細(xì)胞包括激活后的人的T細(xì)胞,自然殺傷性細(xì)胞,肥大細(xì)胞,上皮細(xì)胞,基質(zhì)細(xì)胞,鼠角質(zhì)細(xì)胞,神經(jīng)元和星形膠質(zhì)細(xì)胞。人的IL-3的核苷酸序列是由133個氨基酸組成的多肽序列和19個氨基酸組成的信號序列。在氨基酸水平上,小鼠和人的氨基酸序列只有29%的同源性,雖然同源性比較低,但是在他們在功能中具體種屬特異性。IL-3不僅對造血干細(xì)胞有影響,還可以刺激人的內(nèi)皮細(xì)胞和平滑肌細(xì)胞的增殖和運動。IL-3被證明可以作為神經(jīng)營養(yǎng)因子促進神經(jīng)元的存活和神經(jīng)突的生長。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法。抗人IL-3單抗包被于酶標(biāo)板上,標(biāo)本和標(biāo)準(zhǔn)品中的IL-3會與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人IL-3抗體,生物素化抗人IL-3抗體與酶標(biāo)板上結(jié)合的標(biāo)準(zhǔn)品或樣本中的IL-3結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IL-3,辣根過氧化物酶會使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值,IL-3濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IL-3濃度。
