產(chǎn)品概述
MMP-2(明膠酶 A)在發(fā)育過程中廣泛表達。發(fā)現(xiàn)在組織損傷位點和炎癥發(fā)生位點,MMP-2的表達上調(diào)。轉(zhuǎn)移性腫瘤入侵前緣的基質(zhì)細胞中,MMP-2的表達也上調(diào)。體內(nèi),在腫瘤血管生成和腫瘤轉(zhuǎn)移過程中MMP-2起促進作用。 相比于正常生理狀態(tài),組織損傷、炎癥和癌癥等病理條件下,體液中MMP-2水平升高。人的MMP-2蛋白以一種非糖基化,分子量為72KDa的前體形式被分泌。人MMP-2前體蛋白由兩部分組成:80個氨基酸形成的前體結(jié)構(gòu)域和551氨基酸組成的成熟結(jié)構(gòu)域。人MMP-2前體蛋白的氨基酸序列與小鼠和大鼠約有97%同源性。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??谷薓MP-2單抗包被于酶標(biāo)板上,同時加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗人MMP-2 抗體,標(biāo)本、標(biāo)準(zhǔn)品中的MMP-2會與加入于孔中的生物素化的抗人MMP-2 抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有MMP-2,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值,MMP-2濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中MMP-2濃度。
