產(chǎn)品概述
粒細(xì)胞集落刺激因子(G-CSF)是在嗜中性粒性白細(xì)胞系的造血細(xì)胞的增殖、分化和活化起重要作用的多向性細(xì)胞因子。 G-CSF是通過活化的單核細(xì)胞,巨噬細(xì)胞,成纖維細(xì)胞,內(nèi)皮細(xì)胞,星型膠質(zhì)細(xì)胞,成骨細(xì)胞,骨髓基質(zhì)細(xì)胞等細(xì)胞產(chǎn)生的。除此之外,各種癌細(xì)胞系和成髓細(xì)胞白血病細(xì)胞也能組成性的產(chǎn)生 G-CSF。已經(jīng)表明G-CSF和G-CSF受體能夠通過胎盤,蛻膜和子宮內(nèi)膜腺體細(xì)胞在懷孕期間產(chǎn)生,這表明G-CSF有可能在胎盤和蛻膜中起重要作用。
在人類中,兩個G-CSF的cDNA編碼207和204個氨基酸殘基的前體蛋白質(zhì)已經(jīng)得到了分離。這兩個蛋白質(zhì),除了在成熟蛋白質(zhì)的N端起的35個氨基酸以后的三個氨基酸差異外, 都有一段30個氨基酸的信號肽并且有一致的氨基酸序列。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??谷薌-CSF單抗包被于酶標(biāo)板上, 標(biāo)本和標(biāo)準(zhǔn)品中的G-CSF會與單抗結(jié)合,游離的成分被洗去;加入生物素化的抗人G-CSF抗體,生物素化抗人G-CSF抗體與包被在酶標(biāo)板上的抗人G-CSF單抗結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有G-CSF,辣根過氧化物酶會使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值, G-CSF濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中G-CSF濃度。
