產(chǎn)品概述
白細(xì)胞介素-6是在主體防御中起重要作用的多功能細(xì)胞因子,包括急性感染期、免疫應(yīng)答、神經(jīng)細(xì)胞反應(yīng)和造血作用。IL-6被常規(guī)細(xì)胞和轉(zhuǎn)化淋巴細(xì)胞、非淋巴細(xì)胞所表達(dá)。IL-6的表達(dá)上調(diào)被促有絲分裂信號(hào)、抗原刺激信號(hào)、脂多糖、鈣離子、細(xì)胞因子和病毒等眾多信號(hào)控制。IL-4、IL-10和IL-3調(diào)控IL-6在單核細(xì)胞中的表達(dá)。血清中IL-6水平已經(jīng)應(yīng)用在大量的病理檢測(cè)中,這些檢測(cè)共包括細(xì)菌、病毒感染、外傷、免疫系統(tǒng)疾病和惡性腫瘤。大鼠IL-6 cDNA含211氨基酸殘基的多肽前體蛋白,其具有一個(gè)在成熟蛋白的第187氨基酸殘基位點(diǎn)的狂犬病信號(hào)肽。在蛋白序列水平上,大鼠IL-6和人的同源性約為39%,和小鼠IL-6的同源性為87%。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法。抗大鼠IL-6單抗包被于酶標(biāo)板上,同時(shí)加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗大鼠IL-6抗體,標(biāo)本、標(biāo)準(zhǔn)品中的IL-6會(huì)與加入于孔中的生物素化的抗大鼠IL-6抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IL-6,辣根過氧化物酶會(huì)使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測(cè)OD值, IL-6濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IL-6濃度。
