產(chǎn)品概述
白介素12(IL-12),也稱為自然殺傷細(xì)胞刺激因子(NKSF)或毒性淋巴細(xì)胞成熟因子(CLMF),是一種多功能細(xì)胞因子,在EBV感染的RPMI-8866細(xì)胞培養(yǎng)液中被鑒定出的。IL-12是分子量為75kD的異二聚體組成的糖蛋白,其中,較小的亞單位(p35)與IL-6、G-CSF同源,而大亞單位(p40)被證明與可溶性IL-6受體有同源性。這提示IL-12可能是細(xì)胞因子與IL-6可溶性受體的復(fù)合物進(jìn)化而來的。IL-12具有明顯的種屬特異性,人的IL-12與小鼠系統(tǒng)具有很少的交叉反應(yīng)。巨噬細(xì)胞與B淋巴細(xì)胞表達(dá)的IL-12對T細(xì)胞與自然殺傷細(xì)胞有多種生理效應(yīng)。包括誘導(dǎo)休止期與激活的T細(xì)胞、NK細(xì)胞表達(dá)IFN-γ與TNF,與其他IFN-γ協(xié)同誘導(dǎo)劑在轉(zhuǎn)錄與翻譯水平上誘導(dǎo)IFN-γ表達(dá),增強(qiáng)NK細(xì)胞與T細(xì)胞的殺傷能力,誘導(dǎo)被激活的T細(xì)胞與NK細(xì)胞分裂增殖。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??勾笫驣L-12p70單抗包被于酶標(biāo)板上,同時加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗大鼠IL-12p70抗體,標(biāo)本、標(biāo)準(zhǔn)品中的IL-12p70會與加入于孔中的生物素化的抗大鼠IL-12p70抗體及包被于酶標(biāo)板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IL-12p70,辣根過氧化物酶會使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值,IL-12p70濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IL-12p70濃度。
