產(chǎn)品概述
白介素 2(IL-2)是通過結(jié)合多分子細胞受體復(fù)合物發(fā)揮生物活力的。多年來,這個受體復(fù)合物被認為是由IL-2Rα與IL-2β兩個糖蛋白鏈組成的,兩個亞基相互結(jié)合在一起形成一個高親和力受體來傳遞 IL-2信號的。IL-2Rα(也稱為 Tac 抗原或 CD25),是一個分子量為55kD跨膜糖蛋白,有351個氨基酸組成。IL-2β在1989 年被克隆出來,它是個由575個氨基酸組成的跨膜糖蛋白,分子量為75kD,其中有286個氨基酸位于胞漿內(nèi),更顯著地參與了細胞信號傳導(dǎo)作用。常認為它是IL-2一種弱抑制劑。在任何情況下,體液中增高的可溶性IL-2Rα常伴隨這T細胞與B細胞的增加與免疫系統(tǒng)的激活。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??谷薎L-2 sRα 單抗包被于酶標板上,標本和標準品中的IL-2 sRα 會與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人IL-2 sRα 抗體,生物素化抗人IL-2 sRα 抗體與酶標板上結(jié)合的標準品或樣本中的IL-2 sRα 結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有IL-2 sRα,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值,IL-2 sRα 濃度與OD450值之間呈正相關(guān),可通過繪制標準曲線求出標本中IL-2 sRα 濃度。
